Abstract

An analytical methodology was developed and fully validated to calculate the chronic consume of cocaine, opiates cannabinoids and their metabolites in hair samples, by using d-3,6 MAM-d3 cocaine and THC-COOH-d3 as inner standards. The method comprise a double liquid-liquid extraction, with dichloromethane and Hexane/Ethyl acetate. All substances were detected as their trimethysilyl derivatives (BSTFA/TMCs) in order to increase chromatographic sensitivity and subsequent chromatographic analysis with capillary column HP-5MS de 30 m* 0.25 mm y 0.25 µm. The methodology was selective, linear, precise and accurate. The method was linear over investigated range (one - ten µg/mg) with an excellent intra-assay and inter-assay precision (relative standard deviations, RSD < 15%). Limits of quantification were between 0,1 ng y 7,0 ng/mg and recovery percentages were between 70% y 92%.
